ABSTRACT

Syzygium cumini (L.) Skeels é uma árvore popularmente conhecida como "jamelão" ou "azeitona-roxa", é um vegetal frutífero de grande porte, pertencente à família Myrtaceae e de origem asiática, teve sua expansão em diversas regiões do mundo, no Brasil é encontrada no Nordeste, Norte e Sudeste. Quando aplicada na medicina tradicional apresenta diversas propriedades farmacológicas contidas em suas folhas, frutos, sementes e caule, com isso torna-se uma espécie bastante promissora para a indústria farmacêutica e alimentícia, pois além de conter características benéficas possui um alto poder econômico. Desta forma, este trabalho visa caracterizar físicoquimicamente a droga vegetal e solução extrativa obtidos a partir das partes aéreas de Syzygium cumini (L.) Skeels para futuro desenvolvimento de forma farmacêutica. O material vegetal de Syzygium cumini (L.) Skeels foi coletado na região de Araçoiaba-PE. Suas partes aéreas foram secas e trituradas. A solução extrativa foi obtida através da maceração da matéria-prima utilizando o etanol como solvente e as caracterizações foram realizadas de acordo com a Farmacopeia Brasileira 6° edição.


Syzygium cumini (L.) Skeels is a tree popularly known as "jamelão" or "olive-purple", is a large fruiting plant, belonging to the Myrtaceae family and of Asian origin, had its expansion in several regions of the world, in Brazil is found in the Nor- theast, North and Southeast. When applied in traditional medicine it presents several phar- macological properties contained in its leaves, fruits, seeds and stem, thus becoming a very promising species for the pharmaceutical and food industry, because besides contai- ning beneficial characteristics it has a high economic power. Thus, this work aims to physicochemically characterize the plant drug and extractive solution obtained from the aerial parts of Syzygium cumini (L.) Skeels for future development of pharmaceutical form. The plant material of Syzygium cumini (L.) Skeels was collected in the region of Araçoiaba-PE. Its aerial parts were dried and triturated. The extractive solution was obtained by maceration of the raw material using ethanol as solvent and the characteriza- tions were performed according to the Brazilian Pharmacopeia 6th edition.

Skeels es un árbol conocido popularmente como "ja- melão" o "oliva-púrpura", es una planta fructífera de gran tamaño, perteneciente a la fa- milia Myrtaceae y de origen asiático, tuvo su expansión en varias regiones del mundo, en Brasil se encuentra en el Nordeste, Norte y Sudeste. Cuando se aplica en la medicina tradicional tiene varias propiedades farmacológicas contenidas en sus hojas, frutos, se- millas y tallo, convirtiéndose así en una especie muy prometedora para la industria far- macéutica y alimentaria, ya que además de contener características beneficiosas tiene un alto poder económico. Así, este trabajo tiene como objetivo caracterizar fisicoquímica- mente la droga vegetal y la solución extractiva obtenida de las partes aéreas de Syzygium cumini (L.) Skeels para el futuro desarrollo de forma farmacéutica. El material vegetal de Syzygium cumini (L.) Skeels fue recolectado en la región de Araçoiaba-PE. Sus partes aéreas fueron secadas y trituradas. La solución extractiva se obtuvo por maceración de la materia prima utilizando etanol como solvente y las caracterizaciones se realizaron de acuerdo con la Farmacopea Brasileña 6ª edición.

ABSTRACT

This study determined phytochemical composition, antifungal activity and toxicity in vitro and in vivo of Syzygium cumini leaves extract (Sc). Thus, was characterized by gas chromatography coupled to mass spectrometry and submitted to determination of Minimum Inhibitory (MIC) and Fungicidal concentrations (MFC) on reference and clinical strains of Candida spp. and by growth kinetics assays. Toxicity was verified using in vitro assays of hemolysis, osmotic fragility, oxidant and antioxidant activity in human erythrocytes and by in vivo acute systemic toxicity in Galleria mellonella larvae. Fourteen different compounds were identified in Sc, which showed antifungal activity (MIC between 31.25-125µg/mL) with fungistatic effect on Candida. At antifungal concentrations, it demonstrated low cytotoxicity, antioxidant activity and neglible in vivotoxicity. Thus, Sc demonstrated a promising antifungal potential, with low toxicity, indicating that this extract can be a safe and effective alternative antifungal agent.


Este estudio determinó la composición fitoquímica, la actividad antifúngica y la toxicidad in vitro e in vivo del extracto de hojas de Syzygium cumini (Sc). Así, se caracterizó mediante cromatografía de gases acoplada a espectrometría de masas y se sometió a determinación de Concentraciones Mínimas Inhibitorias (CMI) y Fungicidas (MFC) sobre cepas de referencia y clínicas de Candida spp. y mediante ensayos de cinética de crecimiento. La toxicidad se verificó mediante ensayos in vitro de hemólisis, fragilidad osmótica, actividad oxidante y antioxidante en eritrocitos humanos y por toxicidad sistémica aguda in vivo en larvas de Galleria mellonella. Se identificaron catorce compuestos diferentes en Sc, que mostraron actividad antifúngica (CMI entre 31.25-125 µg/mL) con efecto fungistático sobre Candida. En concentraciones antifúngicas, demostró baja citotoxicidad, actividad antioxidante y toxicidad in vivo insignificante. Por lo tanto, Sc demostró un potencial antifúngico prometedor, con baja toxicidad, lo que indica que este extracto puede ser un agente antifúngico alternativo seguro y eficaz.

ABSTRACT

Syzygiumcumini(L.) Skeels wasadaptedto the climatic conditionsandsoil typesin Brazil. Its fruits, leaves andinner barkare usedin folk medicinedue to their highantioxidant, anti-inflammatory, anticarcinogenicandantidiabeticactivities mainlyassociated with the presenceof phenolic compounds. It is estimated thatat least300million peopleworldwide developdiabetesand approximately 11million peopleare carriersof the disease in Brazil.The objectiveof this workwas to evaluate thein vitro antioxidant activity, as well as thehypoglycemic actionofhydroethanolic extract(HEE), the ethyl acetate(EAF) andhydromethanolic(HMF) fractions from leavesofS.cumini(L.) Skeels in rats. All assays werecarried out in three replications. Data wereexpressed as meanSDand significance was evaluated by ANOVAand Bonferronitest (p < 0.05). The results indicatea significant(p < 0.05) total phenolcontent (207 ± 2.3GAE mg g-1) andantioxidant activity(EC50=9.05±0.170 µg mL-1) for EAF. HEE and its fractions showed no significant (p > 0.05) actionto modulateglucosebytheOGTT assayinnondiabetic micecompared to control. Thus the use of the plant against diabetes in individuals is not proven.

Background: Regulatory guidelines recommend shelf life of herbal products to be established throughsystematic stability studies.Objective: The study was designed to establish shelf life of Syzygium cumini extract through acceleratedand long-term stability testing as per WHO guidelines.Material and methods: The extract was stored under accelerated (40_x005F_x005F_x0001_C/75 %RH) and long-term (25_x005F_x005F_x0001_C/60%RH) stability conditions for 6 and 30 months, respectively. Samples were withdrawn at periodic intervals and analysed through two validated HPLC-UV methods (I and II) for fingerprint and quantitativeanalysis of markers. Antidiabetic activity of control and stability samples was evaluated by a-glucosidaseinhibitory model.Results: Method I generated a well resolved fingerprint of the control sample that was found to containgallic acid (GA, 1.45 % w/w) and ellagic acid (EA, 3.97 % w/w). The content of GA did not change underboth the stability conditions, but that of EA varied insignificantly (3.97e4.77 % w/w) under long-termconditions up to 24 months and subsequently decrease to 3.15 % w/w after 30 months. There was novisible change in LC-UV fingerprint of any stability sample with respect to control. a-Glucosidaseinhibitory activity of all stability samples also remained unaltered as compared to control sample (IC501.48 mg/mL). GA and EA did not elicit any activity at the concentrations present in the extract.Conclusion: Phytochemical composition and antidiabetic efficacy of S. cumini extract remain unchangedduring its storage under both accelerated and long-term stability conditions, which suggest its shelf lifeto be 30 months. Also, GA and EA are not appropriate anti-diabetic markers

ABSTRACT

Syzygium cumini or Eugenia Jambolana Lam is a traditional medicinal plant very native to the Indian and Asian sub-continent. It is commonly known as black plum or jamun. Various plants parts of the tree are known for ethno-medicinal uses, and in particular, the fruits of Jamun tree are well known for medicinal uses and preparation of health drinks. Studies have shown that the berries contain carbohydrates, minerals and the pharmacologically active phytochemicals. The active phytochemical includes flavonoids, terpenes, and anthocyanins. Ayurvedic and Indian Folk Medicine have already mentioned the use of jamun for diabetic treatment even before the advent of insulin. The bark, fruit pulp, seeds and the leaves are evaluated in the crude form as well as extracts in various diabetic models. Further, bioactive phytochemicals are extracted using in vitro assays, some of them such as vitalboside-A, mycominose are also characterized with spectroscopic analysis. This review article is intended to compile Syzygium cumini’s therapeutic potential with special emphasis on the phytochemical constituents and its pharmacological activities related to diabetic intervention through recent studies conducted in last one decade. It also addresses the clinical and toxicological studies.

ABSTRACT

O objetivo do presente trabalho foi desenvolver filmes biodegradáveis antimicrobianos a partir da incorporação do extrato bioativo proveniente da casca do fruto Jamelão. Para tal, foi obtido inicialmente o extrato das cascas em pó e depois foram elaborados filmes biodegradáveis utilizando acetato de celulose e amido+quitosana. Os filmes foram incorporados do extrato em diferentes concentrações para então serem avaliados quanto as suas propriedades antimicrobiana frente às cepas de Staphylococcus aureus e Listeria monocytogenes. Os resultados mostraram que nos filmes de acetato, não houve formação de halo de inibição frente a nenhuma das bactérias testadas, independentemente da concentração de extrato aplicado. Por outro lado, para os filmes de amido com quitosana, foi constatada atividade para as duas bactérias testadas e que à medida que se aumentava a concentração do extrato, aumentavam-se também os tamanhos dos halos de inibição. Logo estes podem ser aplicados em alimentos.

ABSTRACT

The aim of this study was to evaluate the protective effect of three different extracts prepared from Syzygium cumini leaves against paraquat-induced toxicity in Saccharomyces cerevisiaestrains deficient in superoxide dismutase (SOD). Additionally, the extracts phenolic and flavonoid contents, in vitro antioxidant activity, and phytochemical composition (using high-pressure liquid chromatography) were determined. Bioactive compounds from S. cumini leaves were extracted with infusion (traditional method) or ultrasound (aqueous or hydroalcoholic). Compared to the infusion extract, the ultrasound extracts exhibited a greater protective capacity against paraquat toxicity in the yeast cells as well as higher antioxidant activity. These results may be directly related to the higher phenolic and flavonoid contents in these extracts, since they are recognized as having high antioxidant actions.

ABSTRACT

Objective To evaluate the effect of methanolic extract of Syzygium cumini (L.) Skeels (S. cumini) seeds on the major organs in an animal model of diabetes through biochemical and histopathological studies. Methods The methanolic extracts of S. cumini seeds (100 and 200 mg/kg body weight) were administered to alloxan-induced diabetic rats daily, with fasting blood glucose levels being measured by glucometry at one-day interval for a duration of two weeks. Biochemical assays to evaluate changes in the functions of the heart, liver, pancreas and kidney were carried out. Histopathological changes in the diabetic rat organs (pancreas, liver, heart, kidney and spleen) were also observed after the 14 days of treatment with the extracts. Results Oral administration of methanolic extracts of S. cumini seeds (100 and 200 mg/kg body weight), with gliclazide as a positive control (25 mg/kg), showed beneficial effects including lowering blood glucose levels (P < 0.001), improved heart and liver functions, and hyperlipidemia due to diabetes. At 200 mg/kg, the extracts reversed cardiac and liver damage caused by alloxan. Conclusions In addition to the anti-hyperglycemic activity of methanolic extracts of S. cumini seeds, the extracts demonstrates potential to minimize cardiac and hepatic complications.

Objectives: The aim of this study was to identify the slime production and evaluate the effects of Rosmarinus officinalis (rosemary) and Syzygium cumini (jambolan) glycolic extracts, and 0.12% chlorhexidine (CHX) in biofilms formed by strains of coagulase-positive Staphylococcus - CPS and coagulase negative Staphylococcus - CNS isolated from the oral cavity. Material and Methods: Slime production was evaluated by two methods: the color of colony presented in Congo red agar, and through the amount of slime adhered to polystyrene. Biofilms were grown in acrylic resin discs immersed in broth, inoculated with microbial suspension (106 cells/ml) and incubated at 37°C/48 h. After formation, the biofilms were exposed for 5 minutes to glycol extracts, CHX or saline solution. The viability of biofilms was determined by counting the colony-forming units per milliliter (CFU/ml) in agar, and analyzed statistically by Tukey test (p< 0.05). Results: The strains S. aureus, S. schleiferi and S. epidermidis obtained the highest values of slime adhered to polystyrene. R. officinalis promoted reductions ranging from 12.1% to 78.7% in biofilms formed by isolates of CPS, and 9.2% to 73.7% in the biofilms of CNS. S. cumini reduced 12% to 55.7% in biofilms of CPS, and 7.9% to 71.5% in biofilms of CNS. With exception of S. saprophyticus, glycol extracts produced significant reductions in biofilms. For five isolates studied, R. officinalis produced greater reductions than CHX. Conclusion: R. officinalis and S. cumini showed effective antibiofilm activity against isolates that showed slime production.(AU)


Objetivos: O objetivo deste estudo foi identificar a produção de slime e avaliar os efeitos dos extratos glicólicos de Rosmarinus officinalis (alecrim), Syzygium cumini (jambolão) e 0,12% de clorexidina (CLX) em biofilmes formados por cepas de Staphylococcus coagulase positivo (SCP) e Staphylococcus coagulase negativo (SCN) da cavidade oral. Material e Métodos: A produção de slime foi avaliada por dois métodos: a cor da colônia apresentada em ágar vermelho Congo e pela quantidade de slime aderido ao poliestireno. Os biofilmes foram crescidos em discos de resina acrílica imersos em caldo, inoculados com suspensão microbiana (106 células/ml) e incubados a 37°C/48h. Após a formação, os biofilmes foram expostos durante 5 minutos aos extractos glicólicos, CLX ou solução salina. A viabilidade dos biofilmes foi determinada pela contagem das unidades formadoras de colônias por mililitro (UFC/ml) em ágar e analisada estatisticamente pelo teste de Tukey (p< 0,05). Resultados: As cepas S. aureus, S. schleiferi e S. epidermidis obtiveram os maiores valores de aderência ao poliestireno. R. officinalis promoveu reduções variando de 12,1% a 78,7% em biofilmes formados por isolados de SCP e 9,2% a 73,7% nos biofilmes de SCN. S. cumini reduziu de 12% a 55,7% nos biofilmes de SCP, e 7,9% a 71,5% nos biofilmes de SCN. Com exceção de S. saprophyticus, os extratos glicólicos produziram reduções estatísticas nos biofilmes. Para cinco isolados estudados, R. officinalis produziu maiores reduções do que CLX. Conclusão: R. officinalis e S. cumini mostraram atividade antibiofilme efetiva contra isolados que apresentaram produção de slime.(AU)

Traditionally, the Indian Blackberry or locally called Jamun, Eugenia jambolana Lam. (Syn.: Syzygium cumini), is well known for its pharmacological potential, particularly anti-inflammatory. Here, we studied kaempferol-7-O-α-L-rhamnopyranoside]-4'-O-4'- [kaempferol-7-O-α-L-rhamnopyranoside (EJ-01) isolated from the E. jambolana leaves for possible anti-inflammatory activity. EJ-01 (3, 10 and 30 mg/kg, p.o.) was assessed for anti-inflammatory activity using carrageenan-induced paw edema model in mice by determining edema volume, myeloperoxidase (MPO), nitrite plus nitrate (NOx) and cytokine levels in paw edema tissue. EJ-01 significantly attenuated the edema, MPO levels, tumor necrosis factor-alpha (TNF-α) and interleukin-1beta (IL-1β) levels in the edema of paw at the 5th hour after carrageenan injection at all doses. EJ-01 (30 mg/kg) decreased the nitric oxide (NO) levels of the edema of paw at the 5th hour after carrageenan injection. The anti-inflammatory mechanisms of EJ-01 might be related to the decrease in the level of edema paw by reduced activities of NO and MPO. It probably exerts anti-inflammatory effects through the suppression of TNF-α and IL-1β. Therefore, we conclude that EJ-01 could be positively exploited for itspotential benefits against inflammatory diseases and support the pharmacological basis of E. jambolana as traditional herbal medicine for the treatment of inflammatory diseases.

Abstract The fungus Beauveria bassiana is naturally found in poultry houses and causes high rates of mortality in Alphitobius diaperinus. Laboratory and field experiments have shown the potential of this fungus as an insect control agent. However, in poultry houses, bacteria as Salmonella, can be found and have been studied alternative control methods for this pathogen. Thus, this study aimed to evaluate the effect of plant extracts and a disinfectant on the fungus Beauveria bassiana (strain Unioeste 4). Conidial viability, colony-forming unit (CFU) counts, vegetative growth, conidia production, insecticidal activity of the fungus and compatibility were used as parameters in the evaluation of the effect of these products on the fungus. Alcoholic and aqueous extracts of jabuticaba (Myrciaria cauliflora (Mart.), guava (Psidium guajava (L.)), and jambolan (Syzygium cumini (L.), at concentrations of 10% as well as the commercial disinfectant, Peroxitane® 1512 AL, were evaluated at the recommended concentrations (RC), 1:200 (RC), 0.5 RC and 2 RC. There was a negative influence of alcoholic and aqueous extracts of jabuticaba, guava and three dilutions of Peroxitane on the viability of conidia. The CFUs and vegetative growth of the fungus were affected only by the Peroxitane (all dilutions). For conidial production, the aqueous extract of guava had a positive effect, increasing production, while the Peroxitane at the R and RC concentrations resulted in a negative influence. The mortality of A. diaperinus, caused by the fungus after exposure to these products, was 60% for the peracetic acid at 0.5 RC, and above 80% for the extracts. Thus, the results showed that all the extracts and Peroxitane at RC 0.5 are compatible with the fungus B. bassiana Unioeste 4, however only the extracts had a low impact on inoculum potential.


Resumo O fungo Beauveria bassiana é encontrado naturalmente em aviários de frango de corte, tendo sua eficácia como agente controlador do Alphitobius diaperinus, em condições de laboratório e campo. No entanto, nos aviários encontram-se também bactérias, como a Salmonella, para a qual vêm sendo pesquisadas alternativas de controle. Sendo assim, o objetivo do trabalho foi avaliar o efeito de extratos vegetais e um desinfetante com potencial de uso contra Salmonella spp., sobre os parâmetros biológicos do fungo B. bassiana isolado Unioeste 4. Foram avaliados extratos alcoólicos e aquosos de folhas de jabuticabeira (Myrciaria cauliflora (Mart.)), goiabeira (Psidium guajava (L.)), jamboleiro (Syzygium cumini (L.)), na concentração de 10% e também o desinfetante comercial Peroxitane®1512 AL na concentração recomendada – 1:200 (CR), 0,5 CR e 2CR. Foram avaliados a: germinação dos conídios, unidades formadoras de colônias (UFC), crescimento vegetativo, produção de conídios e efeito sobre a atividade inseticida do fungo contra adultos de A. diaperinus, bem como a compatibilidade entre produtos e o fungo. Verificou-se influência negativa dos extratos alcoólico e aquoso de jabuticabeira, goiabeira e das três diluições de Peroxitane sobre a viabilidade dos conídios. Já, a UFC e o crescimento vegetativo foram afetados apenas com Peroxitane (em todas as diluições). Para produção de conídios, o extrato aquoso de goiabeira teve efeito positivo, elevando a produção, enquanto as diluições recomendada e o dobro de Peroxitane mostraram influência negativa. Observou-se ainda que a mortalidade de A. diaperinus causada pelo fungo após a exposição aos produtos foi de 60% para o ácido peracético na 0,5 CR, já para os extratos foi acima de 80%. Assim, os resultados demonstraram que todos os extratos e o Peroxitane na 0,5 CR são compatíveis com o fungo B. bassiana Unioeste 4, porém apenas os extratos tiveram baixo impacto sobre o potencial de inóculo do fungo.

ABSTRACT

Introducción: la especie Syzygium cumini (L.) Skeels, (jambolam), sin. Eugenia jambolana L, es una planta rica en metabolitos secundarios con un elevado potencial biológico, medicinal, entre otros y que pueden estar asociados con las propiedades terapéuticas reportadas para S. cumini. Objetivo: comparar capacidad antioxidante de extractos del fruto, pulpa y semilla de S. cumini, con respecto al contenido de polifenoles y antocianinas. Métodos: fruto, pulpa y semillas S. cumini deshidratados y macerados se sometieron a extracción sólido-líquido; el contenido de fenoles se evaluó por el método de Folin-Ciocalteu, el de antocianinas por el método de diferencial de pH y el potencial antioxidante in vitro se evaluó con base en los métodos del radical estable 1,1-difenil­2-picrilhidracil, el radical catión 2,2´-azino-bis(3-etilbenzotiazolin-6-sulphonic ácido) y poder antioxidante de reducción del ion férrico. Resultados: el contenido de fenoles obtenidos tanto en los extractos metanólicos como etanólicos fue muy similar en fruto y pulpa, mientras que en las semillas se observó un incremento hasta 5 veces con respecto a la pulpa, presentó correlación con el efecto de protección antioxidante entre los extractos evaluados. Conclusiones: la elevada presencia de fenoles determinada en todos los extractos afectó de manera positiva la capacidad protectora contra radicales libres, y los frutos de S. cumini, podrían ser considerados como una fuente potencial de compuestos biaoctivos con aplicaciones terapéuticas(AU)


Introduction: Syzygium cumini (L.) Skeels, (jambolam), sin. Eugenia jambolana L, is considered as a source of secondary metabolites with a biological and medicinal potential and these can be associated with its therapeutics properties. Objetive: to correlated the content of polyphenols and anthocyanins with antioxidant capacity in ethanolic and methanolic extracts of S. cumini. Methods: dehydrated and powered fruits, pulp and seeds of S. cumini was subjected to solid-liquid extraction. Total phenolis content was determined by Folin-Cicoulteau method, anthocyanin content was determined by the pH differential method and the potential antioxidant was evaluated using 1,1-difenil­2-picrilhidracil radical stable, 2,2´-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical cation and ferric reducing/antioxidant power tests. Results: all extracts showed an important correlation between total phenolis content and antioxidant capacity values, however, in the seed total phenolis content was almost 5 times higher than that obtained in fruit and pulp. Conclusions: all extracts exhibited a considerable antioxidant potential against free radical, and S. cumini could be used as an alternative source of natural bioactive compounds with application in therapeutic research area(AU)

ABSTRACT

RESUMO: A espécie Syzygium cumini conhecida como Jambolão é uma das plantas mais utilizadas na medicina popular principalmente no tratamento de febre, estomalgia, gastropatia e diabetes mellitus tipo 2. O rendimento extrativo de compostos bioativos, e mesmo sua propriedade farmacológica, no caso, a capacidade de desativação de radicais livres, são influenciados pelo método e solvente utilizados. No presente estudo foi avaliado o efeito de extração aquosa a quente e a frio, e as extrações hidroalcoólicas em quatro diferentes concentrações de etanol, de folhas e frutos de Jambolão em diferentes estágios de maturação. A quantificação de compostos fenólicos dos extratos foi realizada pelo método de Folin-Ciocalteau, enquanto o método para avaliar a atividade antirradical livre foi realizado através da capacidade de desativação do radical DPPH●. As folhas apresentaram maior quantidade de compostos fenólicos totais (237,52 mg. EAG100 g-1) em relação a qualquer dos cinco extratos dos frutos, independente do estágio de maturação destes. Entre os extratos dos frutos, aquele proveniente de frutos verdes apresentou maior quantidade de compostos fenólicos (109,17 mg.EAG100 g-1). A extração hidro-etanólica a 50% de etanol promoveu os maiores conteúdos de fenóis totais, independente do material vegetal. A maior atividade antirradical livre encontrada dentre os extratos mais ricos em fenóis foi obtida com frutos verdes, IC50 = 2,27 mg.mL-1, a partir da infusão a quente por 30 minutos. Nos extratos de folhas, a maior atividade antirradical livre encontrada foi IC50= 23,07 mg.mL-1 proveniente do extrato hidro-etanólico a 50 % (v/v)


ABSTRACT: Determination of phenolic compounds and antioxidant capacity of aqueous and ethanolic extracts of Jambul (Syzygium cumini). The species Syzygium cumini, known as Jambul, is one of the most used species in folk medicine because of the possible anti-inflammatory and hypoglycemic activity from its extracts. Studies looking for bioactive compounds in several species have tested many extraction methods evaluating the capacity of sequestration of different types of compounds. In the present study, the effects of hot and cold extraction, as well as hydroalcoholic extraction at four different concentrations were evaluated, using as plant material leaves and fruits at different stages of maturation. The content of phenolic compounds of the obtained extracts was evaluated by the Folin-Ciocalteu method, as well as the antiradical activity by the DPPH● scavenging capacity. Analyzing the five different stages of fruit ripening compared to leaves, it was observed that the leaves had higher averages in the amounts of phenolic compounds (237.52 mg GAE 100 g-1) compared to the fruit, being the greenest fruit the one that had the highest amount of phenolic compounds (109.17 mg GAE 100 g-1) when compared to the other stages of maturation tested. The extraction using 50% ethanol showed the best phenol results regardless of the plant material used. The antioxidant activity in the best results for phenols showed a higher antioxidant activity for green fruits (IC50 2.27 mg.mL-1) compared to the leaves (IC50 23.07 mg.mL-1) regardless of the extract type used

ABSTRACT

RESUMO Este estudo teve como objetivo determinar o perfil fitoquímico e avaliar a atividade antimicrobiana in vitro do extrato etanólico da casca do caule de Syzygium cumini(L.) Skeels frente a microrganismos bucais. O perfil fitoquímico do extrato foi traçado através da determinação espectrofotométrica quantitativa para verificar o teor de taninos, flavonóides, saponinas e polifenóis. A atividade antimicrobiana foi determinada através da Concentração Inibitória Mínima (CIM), por meio da técnica de microdiluição em caldo, utilizando-se as seguintes linhagens de microrganismos: Streptococcus mutans (25175), Streptococcus oralis (10557) e Candida albicans (10231). Uma quantidade apreciável de fitocontituintes foi observada, especialmente de taninos (100,58 ± 1,81). Os extratos apresentaram atividade antimicrobiana inibindo o crescimento das linhagens em estudo, destacando-se essa atividade sobre o crescimento de C. albicans (CIM=250 µg/mL). Já as CIMs para Streptococcus foram baixas. Diante dos resultados expostos, pode-se concluir que o perfil fitoquímico foi traçado e que, dentre os microrganismos testados, o extrato etanólico da casca de S. cumini apresentou forte potencial de inibição sobre o crescimento de C. albicans e fraca inibição frente aos Streptococcus testados. Este estudo sugere que mais pesquisas devem ser realizadas dando continuidade à bioprospecção, por meio de análises experimentais com essa espécie vegetal, objetivando, no futuro, que essa planta possa ser utilizada clinicamente para tratar candidose bucal.


ABSTRACT This study aimed to determine the phytochemical profile and to evaluate the in vitro antimicrobial activity of the ethanol extract of stem bark of Syzygium cumini (L.) Skeels against oral microorganisms. The phytochemicalprofile of the extract was traced through a quantitative spectrophotometric determination in order to check the tannin, flavonoids, saponins, and polyphenols content. The antimicrobial activity was determined through minimum inhibitory concentration (MIC) by the broth microdilution technique, using the following strains of microorganisms: Streptococcus mutans (25175), Streptococcus oralis (10557) and Candida albicans (10231). An appreciable amount of fitocontituintes was observed, particularly the tannin (100.58 ± 1.81). The extracts showed antimicrobial activity, inhibiting the growth of the strains under study, with this activity being more intense on the growth of C. albicans ( MIC = 250 mg / mL). On the other hand, the MICs of the Streptococcus were low. In face of the mentioned results, we can conclude that the phytochemical profile was traced and that, among the tested microorganisms, the ethanol extract of S. cumini bark showed strong potential to inhibit the growth of C. albicans and weak inhibition against the Streptococcus tested. This study suggests that more research should be done by proceeding with the bioprospecting, through experimental tests with this plant`s species, aiming that in the future this substance can be used clinically for the treatment of oral candidiasis.

ABSTRACT

Myrica esculenta (Myricaceae) and Syzygium cumini(Myrtaceae), the Indian traditional fruits were studied for their biochemical assay, antifungal activity and mineral analysis. The biochemical assay in terms of total phenols, flavonols, ortho dihydroxy phenolic, ascorbic acid & phytic acid content and antioxidant activity revealed different quantitative make up in all the different samples. The in vitro antioxidant activity determined by reducing power assay, chelating power on Fe2+ ions and 2, 2-diphenylpicrylhydrazil (DPPH) radical scavenging activity were found to be good to moderate in a dose dependent manner in all the extracts. The extracts showed significant antinociceptive and antipyretic activity on mice. Methanolic extracts of both the fruits collected from different regions exhibited good antifungal activity against the three pathogenic fungi viz; Rhizoctonia solani, Sclerotium rolfsii, and Fusarium oxysporium. All the extracts studied for their mineral analysis through atomic absorption spectroscopy also revealed quantitative variation of N, P , K , S, Fe, Zn, Cu and Mn. Fe was found to be maximum in all the samples however Na was absent in all the samples.

Introducción: la aparición de cepas bacterianas resistentes a los antibióticos ha estimulado el desarrollo de nuevas líneas de investigación, basadas en las propiedades antibacterianas de plantas utilizadas en medicina popular. La medicina veterinaria ha estado tomando para sí los beneficios de la medicina tradicional, en un intento de sustituir el uso de antibióticos en el tratamiento y la prevención de la mastitis. Objetivo: evaluar la actividad antibacteriana de los extractos hidralcohólicos de Syzygium cumini (L.) Skeels, en comparación con las bacterias relacionadas en la mastitis bovina. Métodos: se obtuvieron extractos de hojas frescas con alcohol a 92,8° y de hojas secas con alcohol a 70°. La actividad antimicrobiana de plantas se evaluó utilizando la técnica de microdilución. Resultados: los extractos de hojas frescas mostraron una actividad mejor en comparación con los de hojas secas, con valores de concentración mínima inhibitoria de 1,5 a 50 %, en comparación con los diferentes microorganismos. Se destaca como más eficaz contra todo estudio el Streptococcus, con la concentración mínima inhibitoria igual a 3,1; 1,5 y 1,9 % para Streptococcus uberis, Streptococcus agalacteae y Streptococcus dysgalacteae, respectivamente. Conclusiones: estos resultados sugieren que el uso de hojas de esta especie de planta puede constituir una alternativa viable para la prevención y el tratamiento de la mastitis bovina.


Introduction: the emergence of bacterial strains resistant to antibiotics has stimulated the development of new lines of research based on the antibacterial properties of plants used in folk medicine. Veterinary medicine has been taking for itself the benefits of traditional medicine in an attempt to replace the use of antibiotics in the treatment and prevention of mastitis. Objective: this study is aimed to evaluate the antibacterial activity of hydroalcoholic extracts Syzygium cumini (L.) Skeels, compared to bacteria related to bovine mastitis. Methods: the extracts were obtained from fresh leaves with ethanol at 92.8° and leaves with ethanol at 70°. The antimicrobial activity of plants was evaluated using the microdilution technique. Results: the hydroalcoholic extracts fresh plant showed better activity compared with the hydroalcoholic extracts dry leaf, with minimal inhibitory concentration values ranging from 1.5 to 50 % compared to the different microorganisms; highlighting Streptococcus as more effective against all study, with minimal inhibitory concentration equal to 3.1, 1.5, and 1.9 % for Streptococcus uberis, Streptococcus agalacteae, and Streptococcus dysgalacteae, respectively. Conclusions: these results suggest that the use of leaves of this plant species may constitute a viable alternative for the prevention and treatment of bovine mastitis.

ABSTRACT

The anti-inflammatory and apoptotic activity of the essential oil of Syzygium cumini (L.) Skeels, Myrtaceae, leaves was investigated in vivo. The anti-inflammatory action and chronic granulomatous inflammation in BALB/c mice, intravenously infected with Mycobacterium bovis, BCG, (Bacillo Calmet Guerim), was judged by measuring and classifying the granulomas formed in the hepatic parenchyma. The degree of apoptosis in the inflammatory cells was also measured. A reduction in the granulomatous area and a change in the pattern of the granulomas were found. Anti-mycobacterial activity of the essential oil against M. bovis was detected in vitro by an interferometric method in liquid culture medium. The chemical constituents of the essential oil were determined by GC/MS. Higher yields of the essential oil of S. cumini leaves were obtained by extraction in a Clevenger apparatus when the fresh leaves were previously frozen as a pre-processing step. The essential oil obtained from this plant demonstrated a statistically significant and dramatic effect in the chosen model system.

ABSTRACT

Traditionally the leaves of Syzygium cumini (Myrtaceae) are widely used for treating diabetes. The present study was carried out to identify the putative antidiabetic constituents from the S. cumini leaves. From the NMR data four different compounds, Lupeol, 12-oleanen-3-ol-3ß-acetate, Stigmasterol, ßsitosterol were identified from n-hexane fraction of plant extract. These compounds have potential antidiabetic activities which support the traditional use of the leaves as being remedy for treating diabetes.

Syzygium cumini Linn. (Myrtaceae) is a medicinal tree (Jamun) used worldwide in treatment of diabetes. However, no molecular data is available on genetic polymorphism and its relationship, if any with fluoride pollution. In the present study, the genetic variability of two populations of S. cumini growing in fluoride rich soils and normal soils located in Rajasthan and Haryana regions of India, respectively was determined using random amplified polymorphic DNA (RAPD) markers. Different measures of diversity in Rajasthan populations: Shannon’s index of phenotypic diversity (I) = 0.440; Nei’s genetic diversity (h) = 0.292; effective number of alleles per locus (Ne) = 1.497; total species diversity (Hsp) = 0.307 and within population diversity (Hpop) = 0.158 showed high diversity in comparison to Haryana populations. Thus, it seems that Rajasthan population responds with increased genetic variation resulting possibly from new mutation that affect allele frequencies as a consequence of adaptation to contaminated environment. This may imply that the increased diversity levels may act as a buffer to combat fluoride stress. Cluster analysis and principal component analysis (PCA) results showed mixing between the populations.
